چکیده

Type 1 diabetes is characterized by an immune-mediated destruction of the beta-cells, associated with autoantibodies against islet antigens developing years ahead of the clinical manifestation. However, the disease is heterogeneous, and its pathogenesis is far from fully understood. Besides focus on the importance of virus, hygiene, early nutrition, gut microbiome, etc., one hypothesis is that factors leading to beta-cell stress, usually associated with type 2 diabetes, also might contribute to development of type 1 diabetes (1). Islet amyloid is to a varying degree present in almost all individuals with type 2 diabetes, and the beta-cell product islet amyloid polypeptide (IAPP) is the primary constituent of the islet amyloid. IAPP is a member of the calcitonin gene

Islet Amyloid Polypeptide is not a Target Antigen for CD8+ T-Cells in Type 2 Diabetes

Background: Type 2 diabetes (T2D) is a chronic metabolic disorder in which beta-cells are destroyed. The islet amyloid polypeptide (IAPP) produced by beta-cells has been reported to influence beta-cell destruction. Objective: To evaluate if IAPP can act as an autoantigen and therefore, to see if CD8 + T-cells specific for this protein might be present in T2D patients. Methods: Peripheral blood ...

Expression of Interferon-Stimulated Genes in Insulitic Pancreatic Islets of Patients Recently Diagnosed With Type 1 Diabetes.

A primary insult to the pancreatic islets of Langerhans, leading to the activation of innate immunity, has been suggested as an important step in the inflammatory process in type 1 diabetes (T1D). The aim of this study was to examine whether interferon (IFN)-stimulated genes (ISGs) are overexpressed in human T1D islets affected with insulitis. By using laser capture microdissection and a quanti...

Digital Comprehensive Summaries of Uppsala Dissertations from the Faculty of Medicine 1263. 53 pp. Uppsala: Acta Universitatis Upsaliensis. ISBN 978-91-554-9711-8. Type 1 diabetes (T1D) results from a loss of functional insulin-producing pancreatic beta cells. The etiology of T1D is poorly understood, but the detection of infi...

High Plasma Levels of Islet Amyloid Polypeptide in Young with New-Onset of Type 1 Diabetes Mellitus

AIMS/HYPOTHESIS Islet amyloid polypeptide (IAPP) is a beta cell hormone secreted together with insulin upon glucose stimulation. IAPP participates in normal glucose regulation, but IAPP is also known for its ability to misfold and form islet amyloid. Amyloid fibrils form through smaller cell toxic intermediates and deposited amyloid disrupts normal islet architecture. Even though IAPP and amylo...

Function of Isolated Pancreatic Islets From Patients at Onset of Type 1 Diabetes: Insulin Secretion Can Be Restored After Some Days in a Nondiabetogenic Environment In Vitro: Results From the DiViD Study.

The understanding of the etiology of type 1 diabetes (T1D) remains limited. One objective of the Diabetes Virus Detection (DiViD) study was to collect pancreatic tissue from living subjects shortly after the diagnosis of T1D. Here we report the insulin secretion ability by in vitro glucose perifusion and explore the expression of insulin pathway genes in isolated islets of Langerhans from these...
